They figure out how many calories in a particular food, by putting it into a device called a "Bomb calorimeter". What we call a calorie is actually called a kilocalorie, which is the energy needed to increase the temperature of 1 kg of water by 1 degree Celcius. The bomb calorimeter measures how many kilocalories are in a particular type of food.

The way they figure out the calories in a particular type of food is usually done by summing up the calories of all the ingredients.

They determine serving size by what a reasonable amount of in one serving. So nowadays, a can of soda is one serving, since the average person drinks a full can. A small bag of chips is one serving, since one person consumes the complete small bag with their lunch.

The suggestions are based on a 2,000 calorie a day diet. When they calculate the percentage of your daily allowance, its how many calories of the food item as a percentage of that 2,000 calories.
