Canned vegetables are cooked, so depending on how the tomatillos are being used in the recipe you probably want to cook them somewhat before adding them. Also as someone else said if the recipe wants the juice from the can as well, cooking them will give you the liquid you need. I would recommend peeling the leaves off, placing 6-7 fresh tomatillos in a shallow pan, add water to just cover, salt and bring to a boil. Simmer until they can be easily pierced with a fork. Remove from heat. Pour off all but about 6 ounces of water and reserve. A standard can is about 14 ounces, so prep the tomatillos as you need them for the recipe (probably diced) put in a 2 cup measuring cup along with any liquid that comes out on the cutting board and add the reserved water to equal about 14 oz.

Probably, but if it calls for the WHOLE can, juice and all, you might want to cut them up, add some H2O and let them sit and stew in the fridge for a day.
Your recipe might need the juice for flavoring as well as for the liquid.
