Depending on who's making them...Americans tend to change traditional Italian recipes a lot. Stromboli is actually a stuffed and rolled sandwich that is baked. It can contain any fillings, but traditionally meats, vegies and cheeses. Calzone is like a pizza that gets filled and folded over and sealed and then baked. Can be filled with anything-meats, cheese, vegies. Now Panzerotti is a calzone that is deep fried, not baked. Confusing, I know, but they are each delicious!
Yes, it's in the fillings. Strombolis normally include meats and more veggie type fillings, maybe olives and peppers.
Calzones are usually just mozarella and ricotta cheese and sauce.

Another major difference is that calzones are rolled out like a pizza, stuffed on one half, and folded in half.
Stromboli is also rolled out, but in a rectangle, everything is layed out on it, and it gets rolled up like a giant cinnamon bun.
If you see a stromboli made a calzone (done in a lot of restaurants), they are not being made by Italians and most likely have no idea what a stromboli is.
When I make a stromboli, it's like a large loaf of pizza bread which is cut into small slices after it's baked. People invited to parties at my house ALWAYS ask if I'm making stromboli. Calzones are more like pizzas folded over like a turnover and an individual one is made for each person.
